If we have a networked player that has a 16GB SD card, and is currently playing a presentation that totals 8GB in size, what happens if we upload a 12GB presentation?

The publish will fail, or in the case of Simple File Networking or BrightSign Network, it will never complete downloading.
The new presentation content must be downloaded in its entirety before the player will switch over, so there needs to be enough space for both the current scheduled content as well as the new schedule's content.
To work around this, publish a small presentation such as a company logo, let that download, then publish the large update. Once the small presentation is playing, the space occupied by the previous presentation will be available to be freed and used when the large update is published.

Not exactly. If a piece of content is used in an active presentation, then it won't be overwritten.
